Microsoft supports using Data Deduplication with DFS Replication. Optimizing or reversing optimization does not itself trigger replication because the file has not changed. DFS Replication uses Remote Differential Compression for transmission savings rather than transferring the deduplication chunk store. A replica can be optimized independently when deduplication is enabled there. For clustered deduplicated volumes, scheduled deduplication jobs follow the volume and run at the next scheduled interval after another node takes ownership. Microsoft documentation.

Use this review when combining deduplication with a replicated or clustered file service. Identify which members actually use deduplication and which process is responsible for the operation under investigation.

Keep separate operational records for file replication and storage optimization. Ask the storage owner to record job schedules and the replication owner to define a controlled file-change test. Investigate unexpected traffic against actual file changes before attributing it to optimization. Include the destination configuration in the review instead of assuming that it matches the source.
Verification
Observe a permitted optimization operation and a separate intentional file change. Record the relevant replication and optimization results with timestamps. For a clustered volume, include ownership and scheduled-job state around an approved move. Compare the evidence with the expected independent behaviors and investigate discrepancies before broadening the configuration.
